There is no consistency in our settings pages in how preferences are applied:

User profile: Click "Update" to apply changes
UI theme, editor theme: Click "Apply" to apply changes
Plugins, Formatting, General, Validation: Changes to effect immediately with no button push

This is maybe a short topic for a future UX call to come up with consistent behaviour here. Personally I find changes taking effect automatically is ok, although a subtle "Changes have been saved" message appearing somewhere would be comforting. Once preferences become really complicated, ability to cancel changes or restore defaults is valuable, but I don't think we're at that stage.
